How It Works

Phosphate Assay Schematic - PNP Method

Our proven colorimetric method produces a purine product that increases absorbance at 360 nm in direct proportion to inorganic phosphate concentration. Simply measure your samples against the included phosphate standards and calculate concentrations using linear regression—it's that straightforward.

Materials You Will Need

You will need a graduated cylinder, variable pipetters, a test tube vortex-type mixer, spectrophotometer capable of measuring 360 nm with UV compatible cuvettes, clean and phosphate-free 13 x 100mm test tubes, a timer, HPLC water, and ice.

Kit Contents

  • MESG in dry powder form
  • Purine Nucleoside Phosphorylase (PNP) freeze-dried protein glass
  • HEPES/MgCl2 buffer
  • Phosphate Standard (100 ppm Phosphate) in liquid form
  • Standard tubes – 5 tubes (1.5 mL) for preparing Phosphate Standards
