Use this kit for quantification of nitrate in essentially any aqueous sample. Reactions occur on a 96-well microplate provided with the kit. This test kit is based on the enzyme Nitrate Reductase (NaR), catalyzing the reduction of nitrate to nitrite using the natural electron donor NADH. The nitrite reacts with color reagents under acidic conditions to produce a visible color. The concentration of nitrate in the original sample is determined by measuring absorbance with a microplate reader using a 540 nm filter, versus nitrate concentration in nitrate standards.
This kit is supplied complete with all necessary reagents except for HPLC water. Use this kit for quantitative phosphate analysis of essentially any aqueous sample, from wastewater to soil and plant extracts. Simply extract solid samples using HPLC water before analysis. This test tube format kit is to be used with your UV capable spectrophotometer capable of reading absorbance at 360nm.
Standard Range (5 ppm - 20 ppm Phosphate) or Low Range (0.5 - 5 ppm Phosphate)
Reagents for 25 Samples or 100 Samples
When reaction is complete, the purine product yields an increase in absorbance at 360 nm in equal molar ratio to the inorganic phosphate in the reaction mixture. Using the equation of the linear regression line fitted to the set of phosphate standards, the phosphate concentration a sample can easily be determined.

This kit enables you to prepare extracts of 10 samples and to assay these extracts for NADH: Nitrate Reductase (NaR) activity in roots, stems, leaves, whole plants, fungal or microbial extracts. The enzyme Nitrate Reductase (NaR) catalyzes the reduction of Nitrate to Nitrite using the natural electron donor NADH. The nitrite reacts with color reagents (dyes) under acidic conditions to produce a visible magenta color. The color produced is directly proportional to the nitrate concentration. The quantity of nitrate reduced in the reaction time tells you how much active nitrate reductase is present in your sample.
The Enzyme Commission definition of NaR activity is expressed as micromoles of nitrite produced per minute per ml of enzyme (µmol nitrite formed/ min/ ml of extract), in a buffer at pH 7.5 at 30°C. We provide a Nitrate Reductase Activity Standard (YNaR) at 1.0 Unit per milliliter (1.0 U reduces one µmol nitrate per minute). You will use this YNaR standard to determine the NaR activity in your samples.
